Sean O'Loughlin has been ruled out of England's clash with the Exiles next Friday.
The Wigan captain has already missed the Warriors' last two Super League games after failing to recover from a knee injury.
England coach Steve McNamara has not called up a replacement for the 30-year-old loose forward but opted to choose his line-up from a 19-man squad instead.
England's chief medical officer Dr Chris Brookes said: "Sean has withdrawn from the squad because his recovery from a knee injury has been slower than expected.
"The Wigan medical staff will be liaising with the England medical team about his future progress."
O'Loughlin helped England to victory over France in the final of the 2012 autumn international series and was part of the squad that beat the Exiles in the opening international origin match of 2012 last June.
Competition in the back row is fierce for the game at Warrington's Halliwell Jones Stadium with five players fighting for three spots.
The team begin preparations for the game at Loughborough University on Sunday.
